CD36(ClusterofDifferentiation36)isalsoknownasplateletmembraneglycoproteinIV(GPIV),fattyacidtranslocase(FAT),thrombospondinreceptor,collagenreceptorandscavengerreceptorclassB,member3(SRB3),isamemberoftheclassBscavengerreceptorfamilyofcellsurfaceproteins.ThehumanCD36geneencodesasinglechain472aminoacidresidueproteincontainingbothaN-andaC-terminalcytoplasmictailandanextracellularloop.CD36isfoundonplatelets,erythrocytes,monocytes,differentiatedADIpocytes,mammaryepithelialcells,spleencellsandsomeskinmicrodermalendothelialcells.CD36isamultifunctionalglycoproteinthatactsasreceptorforabroadrangeofligands.Ligandscanbeofprotein-naturelikethrombospondin-1(TSP1),fibronectin,collagensIandIVoramyloid-βaswellasoflipidicnaturesuchasoxidizedlow-densitylipoprotein(oxLDL),anionicphospholipids,long-chainfattyacids(LCFA),highdensitylipoprotein(HDL),phosphatidylserine,apoptoticcellsandbacterialdiacylatedlipopeptides.Theyaregenerallymultivalentandcanthereforeengagemultiplereceptorssimultaneously,theresultingformationofCD36clustersinitiatessignaltransductionandinternalizationofreceptor-ligandcomplexes.ThisinternalizationisindependentofmacropinocytosisandoccursbyanactindependentmechanismrequiringtheactivationSrc-familykinases,JNKandRho-familyGTPases.Cellularresponsestotheseligandsareinvolvedinangiogenesis,inflammatoryresponse(throughTLR4-6andTLR2-6signaling),fattyacidmetabolism,tasteanddietaryfatprocessingintheintestine.
