CD40[TNFRSF5]isamemberoftheTNFreceptorsuperfamilywhicharesingletransmembrane-spanningglycoproteinsandplaysanessentialroleinmediatingabroadvarietyofimmuneandinflammatoryresponsesincludingTcell-dependentimmunoglobulinclassswitching,memoryBcelldevelopment,andgerminalcenterformation.CD40contains4cysteine-richrepeatsintheextracellulardomainandisexpressedinBcells,dendriticcells,macrophages,endothelialcellsandseveraltumorcelllines.ThecognateinteractionbetweenCD40andCD40ligand(CD154)onTcellsactivatesNF-κB,JunN-terminalkinaseandJanuskinasesignaltransducersandactivatorsoftranscriptionpathways.SeveraldifferentTRAFproteins(adapterproteins)havebeenidentifiedtoserveasmediatorsofthesignaltransduction.Inaddition,CD40/CD40Linteractionisfoundtobenecessaryforamyloid-β-inducedmicroglialactivationandthusisthoughttobeanearlyeventinAlzheimerdiseasepathogenesis.DefectsinCD40resultinhyper-IgMimmunodeficiencytype3(HIGM3),anautosomalrecessivedisordercharacterizedbytheinabilityofBcellstoundergoisotypeswitching,aswellasaninabilitytomountanantibody-specificimmuneresponseandalackofgerminalcenterformation.
