Recombinanthumansolublevascularendothelialgrowthfactorreceptor-1(sVEGFR-1)isthenaturallyoccurringformandisaglycosylatedmonomericprotein.ThebiologicalfunctionofsVEGFR-1seemstobeanendogenousregulatorofangiogenesis,bindingVEGFwiththesameaffinityasthefull-lengthreceptor.VEGFR-1isatyrosine-proteinkinasethatactsasacell-surfacereceptorforVEGFA,VEGFBandPGF,andplaysanessentialroleinthedevelopmentofembryonicvasculature,theregulationofangiogenesis,cellsurvival,cellmigration,macrophagefunction,chemotaxisandcancercellinvasion.Itmayplayanessentialroleasanegativeregulatorofembryonicangiogenesisbyinhibitingexcessiveproliferationofendothelialcells.Itcanpromoteendothelialcellproliferation,survivalandangiogenesisinadulthood.Itsfunctioninpromotingcellproliferationseemstobecell-typespecific.PromotesPGF-mediatedproliferationofendothelialcells,proliferationofsometypesofcancercells,butdoesnotpromoteproliferationofnormalfibroblasts(invitro).IthasaveryhighaffinityforVEGFAandrelativelylowproteinkinaseactivity.ItmayfunctionasanegativeregulatorofVEGFAsignalingbylimitingtheamountoffreeVEGFAandpreventingitsbindingtoKDR.ModulatesKDRsignalingbyformingheterodimerswithKDR.Ligandbindingleadstotheactivationofseveralsignalingcascades.ActivationofphospholipaseC-γ(PLCG)leadstotheproductionofthecellularsignalingmoleculesdiacylglycerolandinositol1,4,5-trisphosphateandtheactivationofproteinkinaseC.MediatesphosphorylationofPIK3R1,theregulatorysubunitofphosphatidylinositol3-kinase,leADIngtoactivationofphosphatidylinositolkinaseandthedownstreamsignalingpathway.MediatesactivationofMAPK1/ERK2,MAPK3/ERK1andtheMAPkinasesignalingpathway,aswellasoftheAKT1signalingpathway.PhosphorylatesSRCandYES1andmayalsophosphorylateCBL.
