UbiquitinSpecificPeptidase2(USP2),alsoknownasUBP41andUSP9,isacytoplasmicproteinthatbelongstothepeptidaseC19familyofdeubiquitinatingenzymes.Itiswidelyexpressedandservestodeubiquitinateselecttargetsubstrates.Atleastthreeisoformshavethusfarbeendescribed:USP2a,alsoknownasUSP2-69,USP-41,andUSP2-45.Allisoformscontainthesamecatalyticcore,butshowvariationswithintheirN-andC-terminaldomains.USP2aisthecanonicalisoform.Itis605aminoacids(aa)inlengthandhasapredictedmolecularweightof68kDa.HumanUSP2ashares90%aasequenceidentitywiththemouseandratorthologs.USP-41andUSP-45are362and396aainlength,respectivelyandhavepredictedmolecularweightsof41and45kDa.USP-41andUSP2ahavebeenshowntobecriticalregulatorsofcancerandcellproliferation.Theydeubiquitinatepoly-ubiquitinatedtargetproteins,suchasFAS,MDM2,andCyclinD1,topromoteorblockcancer,dependingonthecontext.USP-45,whoseexpressionappearstoberegulatedbycircADIanandnutritionalcues,isthoughttoregulatehepaticgluconeogenesisandglucosemetabolism.
