Ubiquitincarboxy-terminalhydrolaseL1(UCHL1)isadeubiqutinatingenzyme.UbiquitinUCHL1,alsoknownasPGP9.5,isaproteinof223aminoacidsandoneofthemostabundantproteinsinthebrain(1~2%ofthetotalsolubleprotein).Althoughitwasoriginallycharacterizedasadeubiquitinatingenzymerecentstudiesindicatethatitalsofunctionsasaubiquitin(Ub)ligaseandamono-Ubstabilizer.Alargeamountofmono-UbistightlyassociatedwithUCHL1,inhibitingthedegradationofmono-Ubinthebrain.ThepreciseregulationofUCHL1isessentialforneuronstosurviveandtomaintaintheirproperfunction.UCHL1isinvolvedinthepathogenesisofParkinson’sdisease(PD)andAlzheimer"sdisease(AD).Down-regulationandextensiveoxidativemodificationofUCHL1havebeenobservedinthebrainsofADpatientsaswellasPDpatients.Apost-translationalmodificationofUCHL1thatcontrolsthefunctionofUCHL1ismono-ubiquitination.ItoccursreversIBLytoalysineresidueneartheactivesiteofUCHL1.
