UAF1(USP1-AssociatedFactor1,alsoknownasWDRepeat-containingprotein48orWDR48)isapositiveregulatorofatleastthreedeubiquitinatingenzymesoftheC19peptidasefamilyincludingUSP1,USP12andUSP46.IntheabsenceofUAF1,allthreeofthesedeubiquitinasesarealmostcompletelyinactiveandavailablebiochemicalevidencesuggestsUAF1activationoccursbyincreasingcatalyticturnoverasopposedtoalteredsubstrateaffinity(atleastinthecaseofUAF1/USP1)oftheenzymes.Mostnotably,UAF1stronglyactivatesUSP1deubiquitinationofmonoubiquitinatedFANCD2andPCNA,therebyplayingimportantrolesinDNAdamageresponseandtranslesionsynthesis.TheUAF1/USP12complexhasrecentlybeendescribedasanegativeregulatorofNotchsignaling.DisruptionofUAF1-dependentUSP12activityinterruptsNotchtraffickingtothelysosomes,leADIngtoanincreasedamountofreceptoratthecellsurfaceandtohigherNotchactivity.Atthebiochemicallevel,UAF1/USP12deubiquitinatesthenon-activatedformofNotchincellcultureandinvitro.
