TNFreceptor1[TNF-R1;CD120a]isa55kDatypeItransmembraneproteinmemberoftheTNFreceptorsuperfamilyanditisareceptorforTNF-αandhomotrimericlymphotoxin-α.ItisessentialforproperdevelopmentoflymphnodegerminalcentersandPeyer’spatchesandforcombatingintracellularpathogens.ThroughthebuildingofcomplexesthisreceptorcanactivatethetranscriptionfactorNF-κB,mediateapoptosisandfunctionasaregulatorofinflammation.Anti-apoptoticproteinBCL2-associatedathanogene4(BAG4/SODD)andadapterproteinsTRADDandTRAF2havebeenshowntointeractwiththisreceptorandthusplayregulatoryrolesinthesignaltransductionmediatedbythereceptor.TheadaptermoleculeFADDrecruitscaspase-8totheactivatedreceptor.Theresultingdeath-inducingsignalingcomplex(DISC)performscaspase-8proteolyticactivationwhichinitiatesthesubsequentcascadeofcaspases(aspartate-specificcysteineproteases)mediatingapoptosis.TNF-R1isstoredintheGolgiandtranslocatestothecellsurfacefollowingpro-inflammatorystimuli.TNF-αstabilizesTNF-R1andinducesitssequesteringinlipidrafts,whereitactivatesNF-κBandiscleavedbyADAM-17/TACE.ReleaseofthecleavedTNF-R1ECDhelpstoresolveinflammatoryreactions,sinceitdown-regulatescellsurfaceTNF-R1andprovidessolubleTNF-R1tobindTNF-α.Mutationsinhumanshavebeenshowntobeassociatedwiththetumornecrosisfactorassociatedperiodicsyndrome(TRAPS)orperiodicfeversyndromeandwithseveralneurologicalpathologies.
