TheTIM(Tcell/transmembrane,immunoglobulinandmucin)familyplaysacriticalroleinregulatingimmuneresponses,includingallergy,asthma,transplanttolerance,autoimmunityandtheresponsetoviralinfections.TheuniquestructureofTIMimmunoglobulinvariableregiondomainsallowshighlyspecificrecognitionofphosphatidylserine(PtdSer),exposedonthesurfaceofapoptoticcells.TIM-4(Tcell;immunoglobulin;Mucin-4),alsoknownasSMUCKLER,isa60kDamemberoftheTIMfamilyofimmuneregulatingproteins.TIM-4isexclusivelyexpressedonantigen-presentingcells,whereitmediatesphagocytosisofapoptoticcellsandplaysanimportantroleinmaintainingtolerance.TIM-4bindsspecificallytoTIM-1whichisalsothecellularreceptorforthehepatitisAvirus,andhasbeenimplicatedinthedevelopmentofasthma.Amonghematopoieticcells,TIM-1isexpressedonactivatedBandTcells,preferentiallyintheTh2subsetofCD4+Tcells.TheinteractionofTIM-4withTIM-1inducescostimulatoryandhyperproliferativesignalsinTcells.
