TheTIM(Tcell/transmembrane,immunoglobulinandmucin)familyplaysacriticalroleinregulatingimmuneresponses,includingallergy,asthma,transplanttolerance,autoimmunityandtheresponsetoviralinfections.TheuniquestructureofTIMimmunoglobulinvariableregiondomainsallowshighlyspecificrecognitionofphosphatidylserine(PtdSer),exposedonthesurfaceofapoptoticcells.Tim-3,atypeItransmembraneprotein,containsanimmunoglobulinandamucin-likedomaininitsextracellularportionandatyrosinephosphorylationmotifinitscytoplasmicportion.TIM-3ispreferentiallyexpressedonTh1andTc1cells,andgeneratesaninhibitorysignalresultinginapoptosisofTh1andTc1cells.TIM-3isalsoexpressedonsomedendriticcellsandcanmediatephagocytosisofapoptoticcellsandcross-presentationofantigen.Tim-3functionstoinhibitaggressiveTh1-mediatedauto-andalloimmuneresponses.Tim-3pathwayblockadebyadmiNISTrationofTim-3:Fcfusionproteinacceleratesdiabetesinnonobesediabeticmice,causeshyperproliferationofTh1cellsandTh1cytokinereleaseinanexperimentalautoimmuneencephalomyelitis(EAE)modelandpreventsacquisitionoftransplantationtoleranceinducedbycostimulationblockade.
