TIGIT(TcellimmunoreceptorwithIgandITIMdomains)isanovelmemberoftheimmunoglobulinsuperfamily(IgSF).TIGITandCD226bindthesameligands,thetwonectin-familymemberspoliovirusreceptor(PVR)andpoliovirusreceptor-related2(CD112)andcompeteforeachother.TIGITinhibitswhileCD226activatesthesignalingthroughPVRandCD112.TIGITandCD226directlyinteractatthecellsurfaceandthisinteractionimpairsCD226homodimerizationandfunction.TIGITisabsentonnaiveTcells,butisexpressedonactivatedTregsandmemoryTcells,andonNKcellsandNKTcells.TIGITnegativelyregulatesTcellactivationandblockadeofTIGITexacerbatesseveralimmunediseases.TIGITalsoactivatesthePVRreceptorondendriticcells(DCs).AdditionofTIGIT(human):Fc(human)toDCstriggersphosphorylationofPVRandelicitsdownstreamsignaling,whichtheninhibitsTcellresponsesbyproducingIL-10.TIGITalsonegativelyregulatesNKcellcytotoxicityandcytokineproduction.TIGITisdirectlyregulatedbyFoxP3andpromotesinducIBLeTregdifferentiation.
