Thrombospondin-1(TSP1)isa150-180kDacalcium-sensitiveproteinthatissecretedasadisulfide-linkedhomotrimer.Withinthethrombospondinfamily,TSP-2isalsohomotrimeric,whileTSP-3,-4,and-5/COMParehomopentameric.TSP-1regulatesawiderangeofcellularfunctionsincludingtheirinteractionswithothercellsandwiththeextracellularmatrix(ECM).TSP-1containsanN-terminallamininG-likeglobulardomain,anextendedcentralregionwithoneVWFCdomain,3TSPtype-1domains,2EGF-likedomains,and8TSPtype-3domainsandaglobularTSPC-terminaldomain.DistinctregionsofTSP-1havebeenassociatedwithbindingtoparticularECMorcellularmolecules.TSP-1counteractstheangiogenic,hypotensiveandanti-thromboticeffectsofnitricoxide(NO).ItbindsandneutralizesVEGF,blocksVEGFR-2signalingonvascularendothelialcells(EC)anddestabilizesadhesivecontactsbetweenEC.TSP-1’sactionsonvascularsmoothmusclecellssupportarterialtoneandbloodpressureinthepresenceofvasodilatorystimulation.Itisreleasedfromactivatedplateletsandenhancesplateletaggregationandadhesion.TSP-1alsoplaysanimportantroleinwoundrepairandtissuefibrosisbybindinglatentTGF-βandinducingreleaseoftheactivecytokinefromthelatencyassociatedpeptide(LAP).TSP-1dampensadaptiveimmuneresponsesbyinducingthedifferentiationofregulatoryTcellsandinhibitingTCRsignaling.Inthenervoussystem,itpromotesexcitatorysynapseformationandsupportstheintegrationofneuralProgenitorcellsintotheolfactorybulb.Inaddition,TSP-1isreleasedbyapoptoticcellsandpromotesmacrophage-mediateddebrisclearance.
