Themammalianthioredoxinreductases(TrxRs)areafamilyofselenocysteine-containingpyridinenucleotide-disulfideoxido-reductases.AllthemammalianTrxRsarehomologoustoglutathionereductasewithrespecttoprimarystructureincludingtheconservedredoxcatalyticsite(-Cys-Val-Asn-Val-Gly-Cys-)butdistinctivelywithaC-terminalextensioncontainingacatalyticallyactivepenultimateselenocysteine(SeCys)residueintheconservedsequence(-Gly-Cys-SeCys-Gly).TrxRishomodimericproteininwhicheachmonomerincludesanFADprostheticgroup,aNADPHbindingsiteandaredoxcatalyticsite.ElectronsaretransferredfromNADPHviaFADandtheactive-sitedisulfidetoC-terminalSeCys-containingredoxcenter,whichthenreducesthesubstratelikethioredoxin.ThemembersofTrxRfamilyare55–58kilodaltoninmolecularsizeandcomposedofthreeisoformsincludingcytosolicTrxR1,mitochondrialTrxR2,andTrxR3,knownasTrxandGSSGreductase(TGR).TrxRplaysakeyroleinprotectionofcellsagainstoxidativestressandredox-regulatorymechanismoftranscriptionfactorsandvariousBIOLOGicalphenomena(1).
