Tryptophan2,3-dioxygenase(TDO)isaheme-containingdioxygenasecatalyzingtheadditionofmolecularoxygenacrossthe2,3-doublebondoftheindoleringoftryptophantoformN-formylkynurenine(NFK).InAnophelesgambiae,TDOistheonlyenzymeabletocatalyzethefirstandrate-limitingstepinL-Trpcatabolismthroughthekynureninepathway.Tryptophanisanessentialaminoacidforproteinsynthesisandalsotheprecursorforproductionofanumberofneurotransmitters,suchasSEROtoninandmelatonin;inmosquitoes,thekynureninepathwayisessentialforeyepigmentation.Conceivably,thetryptophan-usingpathwaysshouldberegulatedinacoordinatedmannerinmosquitoesaswellasinotherspeciesandTDOactivation/inactivationprocessescouldplayanessentialroleinthesephenomena.
