Thehumanstromalcell-derivedfactor-1(SDF1),alsoknownasCXCL12,isasmall(8kDa)cytokinethatbelongstotheCXCchemokinesubfamily.SDF1iswidelyexpressedinvariousorgansincludingheart,liver,brain,kidney,skeletalmuscle,andlymphoidorgans,andishighlyconservedbetweenspecies.SDF1isexpressedintwoisoformsfromasinglegenethatencodestwosplicevariants,SDF1αandSDF1β,whichareidenticalexceptforthefourresiduespresentintheC-terminusofSDF1βbutabsentfromSDF1α.Asahighlyefficaciouslymphocytechemoattractant,SDF1activatesanddirectsthemigrationofleukocytesinresponsetoproinflammatorystimulisuchasLPS,TNF,orinterleukins.IthasbeendemonstratedthatSDF1promotestumorgrowthandmalignancy,enhancestumorangiogenesis,participatesintumormetastasis,andthusplaysanimportantroleincarcinomapathogenesis.Inaddition,SDF1isidentifiedastheBIOLOGicalligandforCXC-chemokinereceptor4(CXCR4),whichiswidelyexpressedinleukocytesandactsasthecofactorforHIV-1entry,andinhibitstheHIV-1infection.FurThermore,SDF1israpidlyinactivatedviaN-terminalprocessingbycathepsinGassociatedwiththemembraneofB-cells,NKcellsandtoalesserextent,T-cells.
