S100proteinfamilyconsistsofhighlyhomologousCa2+bindingproteinsthatpossesstwoEF-handmotifs.MostS100proteinsexistasdimers(frequentlyhomodimers)withincells.Exclusivelyexpressedinvertebrates,S100isimplicatedinvariousintracellularandextracellularregulatoryactivities.StudiesindicatethatS100proteinsareinvolvedintheinhibitionofproteinphosphorylation,inhibitionofcytoskeletalconstituentassembly,regulationofCa2+homeostasis,stimulationofenzymeactivities,andinteractionwithtranscriptionfactors.S100Bisabundantinthenervoussystemwhereitispredominantlyexpressedinastrocytes,oligodendrocytesandSchwanncells.Whensecretedbyastrocytes,S100Bhasneurotrophiceffectsduringdevelopmentandnerveregenerationatphysiologic(nanomolar)concentrations.High(micromolar)concentrationofS100B,however,hasshowntobeneurotoxic,participatinginthephysiologyofneurodegenerativedisorders.
