PARP-1(ARTD1)isinvolvedinthebaseexcisionrepair(BER)pathway,bycatalyzingthepoly(ADP-ribosyl)ationofalimitednumberofacceptorproteinsinvolvedinchromatinarchitectureandinDNAmetabolism.ThismodificationfollowsDNAdamagesandappearsasanobligatorystepinadetection/signalingpathwayleADIngtothereparationofDNAstrandbreaks.PARP-1positivelyregulatesthetranscriptionofMTUS1andnegativelyregulatesthetranscriptionofMTUS2/TIP150.ItformsacomplexwithEEF1A1andTXKthatactsasaT-helper1(Th1)cell-specifictranscriptionfactorandbindsthepromoterofIFN-γtodirectlyregulateitstranscription,andisthusinvolvedimportantlyinTh1cytokineproduction.PARP-1(E998Kmutant)isaninactiveformofPARP-1whichcanbeusedasacontrolcompound.
