TheProteasomeActivator28(PA28)RegulatoryComplex,alsoknownasthe11Sregulatory(REG)complex,isaring-shaped,multimeric,ATP-independentregulatorycomplexthatcanbindtooneorbothendsofthe20SProteasomeor20SImmunoproteasome.ThePA28regulatorycomplexstimulatestheproteasometohydrolyzesmallpeptides,butnotubiquitinatedproteins,fortheproductionofpeptidesforMHCclassIantigenpresentation.ThreePA28regulatorycomplexsubunits,α,β,andγ,havebeenidentifiedandcanoligomerizetoformthePA28regulatorycomplex.WhiletheexpressionofthePA28αandβsubunitsareinducedbyIFN-γ,thePA28γsubunitisnotsignificantlyaffected.ThehumanPA28activatorβsubunit,alsoknownas11SREGβ,isa239aminoacid(aa)memberofthePA28familyofregulatoryproteinswithapredictedmolecularweightof27kDa.ThemouseandratPA28βorthologsshare93%and92%aasequenceidentitywiththehumanprotein,respectively.invivo,PA28βformshetero-heptamersor-hexamerswithPA28α.PA28βcannotformactivehomo-oligomersinvitroandcannotactivatethe20SProteasomeintheabsenceofPA28α.InadditiontoregulatingproductionofMHCclassIantigens,thePA28α/βregulatorycomplexmayalsoprotectagainstoxidativestress.
