TheProteasomeActivator28(PA28)RegulatoryComplex,alsoknownasthe11Sregulatory(REG)complex,isaring-shaped,multimeric,ATP-independentregulatorycomplexthatcanbindtooneorbothendsofthe20SProteasomeor20SImmunoproteasome.ThePA28regulatorycomplexstimulatestheproteasometohydrolyzesmallpeptides,butnotubiquitinatedproteins,fortheproductionofpeptidesforMHCclassIantigenpresentation.ThreePA28regulatorycomplexsubunits,α,β,andγ,havebeenidentifiedandcanoligomerizetoformthePA28regulatorycomplex.WhiletheexpressionofthePA28αandβsubunitsareinducedbyIFN-γ,thePA28γsubunitisnotsignificantlyaffected.ThehumanPA28activatorαsubunit,alsoknownas11SREGα,isa249aminoacid(aa)memberofthePA28familyofregulatoryproteinswithapredictedmolecularweightof29kDa.Themouseandratorthologsshare94%and95%aasequenceidentitywiththehumanprotein,respectively.invivo,PA28αformshetero-heptamersor-hexamerswithPA28β.PA28αcanalsoformactivehomo-heptamersinvitroandcanbeusedtoactivatethe20SProteasome.InadditiontoregulatingproductionofMHCclassIantigens,PA28αmayprotectagainstoxidativestress.
