Nucleobindin-2(NUCB2;Prepronesfatin)wasfirstisolatedinADIpocytes,butisalsoexpressedbygastricmucosaandpancreaticβcells.Thispatternofnucleobindin-2expression,aswellasthepresenceofnucleobindin-2withintheplasmaofrodentsandhumans,stronglysuggeststhatthiscompoundmayactasacirculatingregulatoryfactor.NUCB2canbecleavedintothreepolypeptides:nesfatin-1,nesfatin-2,andnesfatin-3.Anorexigeniceffectshavebeendescribedfornesfatin-1thatplaysanimportantroleinhypothalamicpathwaysregulatingfoodintakeandenergyhomeostasisandactinginaleptin-independentmanner.Nesfatin-1mayalsoexerthypertensiverolesandmodulatebloodpressurethroughdirectlyactingonperipheralarterialresistance.Nucleobindin-2isthereforereportedtoreducefoodintakeafteradmiNISTration,mostlyreferingtothecleavedpeptidenesfatin-1.Excessnucleobindin-2/nesfatin-1inthebrainleadstoalossofappetite,lackoftheproteininthebrainleadstoanincreaseofappetite.
