Nicotinamidemononucleotideadenylyltransferase2(NMNAT2)catalyzestheformationofNAD+fromnicotinamidemononucleotide(NMN)andATP.Canalsousethedeamidatedformnicotinicacidmononucleotide(NaMN)asasubstratebutwithlowerefficiency.NMNAT2alsocatalyzesthereversereaction,i.e.thepyrophosphorolyticcleavageofNAD+.Itishighlyexpressedinbrain,inparticularincerebrum,cerebellum,occipitallobe,frontallobe,temporallobeandputamen.NMNAT2isalsofoundintheheart,skeletalmuscle,pancreasandisletsofLangerhans.NMNAT2isessentialforaxongrowthandsurvival.ItslossfrominjuredaxonsmayactivateWalleriandegeneration(axondegenerationinducedbynerveinjury),whereasNMNAToverexpressionrescuesaxonsfromdegeneration.
