Neuregulin-4(Nrg4)belongstoasmallfamilyofEGF-like(EGFL)domain-containingproteinsthataresynthesizedastransmembraneprecursorsandundergoproteolyticcleavage.TheEGF-likedomain(aa5-46)ofNrg4(aa1-53)directlybindstothereceptorsErbB3and4.Nrg4isacoldinducedADIpokine,highlyexpressedinadiposetissuesandenrichedinbrownfat.Itisincreasedduringbrownadipocytedifferentiationandreducedinrodentandhumanobesity.Itpromotesneuriteoutgrowthandprotectsagainstdiet-inducedinsulinresistanceandhepaticsteatosisthroughattenuatinghepaticlipogenicsignaling.ThishepaticeffectofNrg4ismediatedbyErbB3andErbB4signalingthatnegativelyregulatesdenovolipogenesismediatedbyLXRandSREBP1c.ThiseffectofNrg4onfattyliverandinsulinresistancecouldleadtothedevelopmentofNrg4asaneffectivetherapeuticbiologicalforthetreatmentofNAFLDandtype2diabetes.GST-Nrg4(aa1-53)recombinantproteinhasbeenshowntomimictheeffectofendogenoussecretedNrg4onliverlipogenesis.
