Interferon-stimulatedGene15(ISG15),alsoknownasUCRP,isaubiquitin-likeproteinthatiscovalentlycoupledtotargetproteinsinaprocesstermedISGylation.Itisa165aminoacid(aa)polypeptidewithapredictedmolecularweightof18kDa.ISG15exhibits66%aasequenceidentitywithitsmouseortholog.Structurally,ISG15consistsoftwotandemubiquitin-likedomainsthatshareasimilar3-dimensionalstructurewithubiquitinandotherubiquitin-likemodifiersincludingNEDD8andSUMO1.ModificationoftargetsbyISG15occursinastepwiseenzymaticprocesssimilartothatofubiquitin.EnzymesregulatingISGylationincludetheactivating(E1)enzymeUBE1L,theconjugating(E2)enzymeUbcH8,andligases(E3)suchasTRIM25andHERC5.RemovalofISG15iscatalyzedbythedeconjugatingenzymeUBP43/USP18.Functionally,ISG15hasputativerolesintheimmuneresponseandtumOrigenesis.ThisisreflectedbyintracellularISG15targetsthatincludeCyclinD1,tumorsuppressorp63,IRF3andarangeofviralproteins.Itisinducedbytype1interferonsandmicrobialinfectionandknockoutmiceexhibitanincreasedsensitivitytoinfectionbysomeviruses.ISG15canalsobesecretedbycellsoftheimmunesystemandmayactinacytokine-likemanner.Forinstance,itisproducedbyhumangranulocytesinresponsetomycobacteriumexposureandnaturalkillercellsandTcellsrespondtoextracellularISG15withIFN-γproduction.Furthersupportingaroleinimmunefunction,ISG15mutationsareassociatedwithMSMD,aninheriteddisordercharacterizedbyincreasedsusceptibilitytomycobacterialinfection.
