Interleukin-35(IL-35)isanovelIL-12familycytokineproducedbyregulatoryTcells(Treg)butnotbyrestingoractivatedeffectorTcells(Teff).IL-35isaheterodimericproteincomposedofEBI3(Epstein-Barr-Virus-inducedgene3)andIL-12a(p35).EBI3isadownstreamtargetofFoxp3,atranscriptionfactorrequiredforTreg-celldevelopmentandfunction,andthusTreg-cellrestrictionofIL35occurs.RegulatoryTcellsareessentialformaintainingselftoleranceandpreventingautoimmunity,andIL-35isidentifiedasamoleculethatmediatestheimmunesuppressionfunctionofTreg-cell.Asaninhibitorycytokine,IL-35inducesproliferationofTreg-cellpopulationsbutsuppressesTh17celldevelopment.StudiesinmiceshowtheabsenceofeitherIL-35chainfromTreg-cellreducesthecells"abilitytosuppressinflammationusinganexperimentalmodelforinflammatoryboweldisease.IL-35issuggestedasapotentialtargetofimmunotherapy.Recently,insufficientIL-35levelswereshowntoplayapivotalroleinthedevelopmentoftype1diabetes(T1D)andautoimmunediseases.
