Interleukin-27(IL-27)isaheterodimericgroup2receptorligandmoleculethatbelongstotheIL-6/IL-12familyoflongtypeIcytokines.ItiscomposedofEBI3(EBV-inducedgene3),a34kDaglycoproteinthatisrelatedtothep40subunitofIL-12andIL-23,andp28,thecloned28kDaglycoproteinthatisrelatedtothep35chainofIL-12.IL-27isexpressedbymonocytes,endothelialcellsanddendriticcells.IL-27bindstoandsignalsthroughaheterodimericreceptorcomplexcomposedofWSX1(TCCR)andgp130.EvidencesuggestsIL-27interactsonlywithWSX-1.IL-27hasbothanti-andproinflammatoryproperties.Asanantiinflammatory,IL-27seemstoinduceageneralnegativefeedbackprogramthatlimitsTandNK-Tcellactivity.Attheonsetofinfection,IL-27inducesanIL-12receptoronnaïeCD4+Tcells,makingthemsusceptIBLetosubsequentIL-12activity(andpossibleTh1development).Notably,IL-12familycytokinesarebothinducedandinhibitedbybacterialproducts.MicrobespromoteIL-27secretionthroughTLR4,andalsoblockIL-27productionviaC5ainduction.
