Interleukin-21(IL-21)isakeyfactorinthetransitionbetweeninnateandadaptiveimmuneresponsessecretedbyactivatedTcells.TheIL-21receptor(IL-21R)isexpressedinlymphoidtissue,inparticularbyNK,B,Tanddendriticcells,macrophagesandendothelialcells.RecentevidencesuggeststhatIL-21playsasupportiveroleintheproliferationofTandBcellsandinfluencesthecytolyticactivityofnaturalkillercells.IL-21hasbeenshowntoup-regulategenesassociatedwithinnateimmunityandtoinhibitthedifferentiationofnaïveThelpercells.IL-21specificallyinhibitsIFN-γproductionfromdevelopingTH1cellsandispreferentiallyexpressedbyTH2cells.FurThermoreIL-21hasbeenidentifiedasagrowthandsurvivalfactorforhumanmyelomacells.IL-21/IL-21Rinteractionshaveauniqueroleinsequentiallyactivatingbothinnateandadaptiveimmuneresponsesagainstpoorlyimmunogenictumors,leADIngtotumorrejectionthatisperforindependentbutIFN-γindependent.
