Interleukin-2(IL-2)isa133aminoacidglycoproteinwithoneintramoleculardisulfidebondandvariableglycosylation.ItissecretedbyactivatedTcellsandinducesproliferationandmaturationofactivatedTcells,naturalkillercellsandlymphokineactivatedkillercells.IL-2alsostimulatesproliferationofantibody-producingBcells,activatesneutrophilsandinducesmononuclearcellstosecreteIFN-γandTNF-αand-β.Moreover,studieshaveshownthatIL-2isrequiredforactivation-inducedapoptosis,animportanthomeostaticmechanismintheimmunesystem,whichisinvolvedinthemaintenanceofperipheraltolerancetoself-antigens.
IL-2promotesTcellproliferationandparticularlynaiveTcells.IL-2signalingonactivatedTcellsiseffectedthroughaquaternaryhigh-affinityreceptorcomplexconsistingofIL-2,IL-2Rα(CD25),IL-2RβandIL-2Rγ.NaiveTcellsarerelativelyinsensitivetoIL-2astheyonlyexpresssmallamountsofIL-2RβandIL-2Rγ.TheyonlyacquiresensitivityafterCD25expression,whichcapturesthecytokineandpresentsittotheIL-2RβandIL-2Rγreceptors.IL-2Superkine(Fc)isanartificialvariantofIL-2containingmutationsatpositionsL80F/R81D/L85V/I86V/I92F.Thesemutationsarelocatedinthemolecule"scorethatactstostabilizethestructureandtogiveitareceptor-bindingconformationmimickingnativeIL-2boundtoCD25.ThesemutationseffectivelyeliminatethefunctionalrequirementofIL-2forCD25expressionandelicitproliferationofTcells.ComparedtoIL-2,theIL-2superkineinducessuperiorexpansionofcytotoxicTcells,leADIngtoimprovedantitumourresponsesinvivo,andelicitsproportionallylesstoxicitybyloweringtheexpansionofTregulatorycellsandreducingpulmonaryoedema.
