Interleukin-18(IL-18)isacostimulatoryfactorforproductionofinterferon-γ(IFN-γ)inresponsetotoxicshockandsharesfunctionalsimilaritieswithIL-12.IL-18issynthesizedasaprecursor24kDamoleculewithoutasignalpeptideandmustbecleavedtoproduceanactivemolecule.IL-1convertingenzyme(ICE;Caspase-1)cleavespro-IL-18atasparticacidintheP1position,producingthemature,bioactivepeptidethatisreADIlyreleasedfromthecells.ItisreportedthatIL-18isproducedfromKupffercells,activatedmacrophages,keratinocytes,intestinalepithelialcells,osteoblasts,adrenalcortexcellsandmurinediencephalon.IFN-γisproducedbyactivatedTorNKcellsandplayscriticalrolesinthedefenseagainstmicrobiralpathogens.IFN-γactivatesmacrophagesandenhancesNKactivityandBcellmaturation,proliferationandIgsecretion.IFN-γalsoinducesexpressionofMHCclassIandIIantigensandinhibitsosteoclastactivation.IL-18actsonThelpertype-1(Th1)TcellsandincombinationwithIL-12stronglyinducesthemtoproduceIFN-γ.PleiotropiceffectsofIL-18havealsobeenreported,suchasenhancementproductionofIFN-γandGM-CSFinperipheralbloodmononuclearcells,productionofTh1cytokines,IL-2,GM-CSF,IFN-γinTcellsandenhancementofFasligandexpressionbyTh1cells.
