Glutathioneperoxidase4,alsoknownasGPX4,isanenzymethatinhumansisencodedbytheGPX4gene.GPX4isaphospholipidhydroperoxidasethatprotectscellsagainstmembranelipidperoxidation.GPX4catalyzesthereductionofhydrogenperoxide,organichydroperoxidesandlipidperoxidesattheexpenseofreducedglutathioneandfunctionsintheprotectionofcellsagainstoxidativestress.Theoxidizedformofglutathione(glutathionedisulfide),whichisgeneratedduringthereductionofhydroperoxidesbyGPX4,isrecycledbyglutathionereductaseandNADPH/H+.GPX4differsfromtheotherGPXfamilymembersintermsofitsmonomericstructure,alessrestricteddependenceonglutathioneasreducingsubstrate,andtheabilitytoreducelipid-hydroperoxidesinsideBIOLOGicalmembranes.
