Glutathioneperoxidase1alsoknownasGPx-1isanenzymethatinhumansisencodedbytheGPX1gene.Twoalternativelysplicedtranscriptvariantsencodingdistinctisoformshavebeenfoundforthisgene.Thisgeneencodesamemberoftheglutathioneperoxidasefamily,consistingofeightknownglutathioneperoxidases(Gpx1-8)inhumans.MammalianGpx1(thisgene),Gpx2,Gpx3,andGpx4havebeenshowntobeselenium-containingenzymes,whereasGpx6isaselenoproteninhumanswithcysteine-containinghomologuesinrodents.Inselenoproteins,the21staminoacidselenocysteineisinsertedinthenascentpolypeptidechainduringtheprocessoftranslationalrecodingoftheUGAstopcodon.Glutathioneperoxidasefunctionsinthedetoxificationofhydrogenperoxide,andisoneofthemostimportantantioxidantenzymesinhumans.IthasbeenreportedthattheproteinencodedbythisgeneprotectsfromCD95-inducedapoptosisinculturedbreastcancercellsandinhibits5-lipoxygenaseinbloodcells,anditsoverexpressiondelaysendothelialcellgrowthandincreasesresistancetotoxicchallenges.Thisproteinisoneofonlyafewproteinsknowninhighervertebratestocontainselenocysteine,whichoccursattheactivesiteofglutathioneperoxidaseandiscodedbythenonsense(stop)codonTGA.
