Parkinson’sdisease(PD)isthesecondmostcommonneurodegenerativedisorderafterAlzheimer’sdisease,withaprevalenceof1–2%inpopulationaged65yearsorolder.PDisacomplex,multifactorialdisorderthattypicallymanifestsaftertheageof50years.ThepathologyinvolvesthelossofdopaminergicneuronsinthesubstantianigraandthepresenceofLewybodies(intraneuronalaccumulationsofaggregatedproteins),insurvivingneuronsinvariousareasofthebrain.α-synucleinandparkinarethoughttobefirmlyassociatedwithPD.TheexactBIOLOGicalfunctionoftheDJ-1proteiniscurrentlyunknown.Itmayplayaroleintheoxidativestressresponse,andthisfunctioncouldbeimportantinpreventingtheonsetofPD.DefectsinPARK7arethecauseofautosomalrecessiveearly-onsetParkinsondisease7(PARK7).ThehumanDJ-1proteincontains189aminoacidresidues.DJ-1maypossessanactivesiteandcouldalsobeaprotease/hydrolasesimilartothePfpIproteases.TheDJ-1genehasbeenhighlyconservedinevolution.ThestructuresofthemouseandhumanDJ-1genesaresimilar,andhumanandmouseDJ-1proteinsdisplay90%aminoacididentity.
