CD80(B7-1)andCD86(B7-2)togetherwiththeirreceptorsCD28andCTLA-4constituteoneofthedominantcostimulatorypathwaysthatregulateTcellandBcellresponses.AlthoughbothCTLA-4andCD28canbindtothesameligands,CTLA-4bindstoB7-1andB7-2witha20-100foldhigheraffinitythanCD28andisinvolvedinthedown-regulationoftheimmuneresponse.B7-1isexpressedonactivatedBcells,activatedTcells,andmacrophages.B7-2isconstitutivelyexpressedoninterdigitatingdendriticcells,Langerhanscells,peripheralblooddendriticcells,memoryBcellsandgerminalcenterBcells.Additionally,B7-2isexpressedatlowlevelsonmonocytesandcanbeup-regulatedthroughinterferon-γ.B7-1andB7-2arebothmembersoftheimmunoglobulinsuperfamily.IthasbeenobservedthatbothhumanandmouseB7-1andB7-2canbindtoeitherhumanormouseCD28andCTLA-4,suggestingthatthereareconservedaminoacidswhichformtheB7-1/B7-2/CD28/CTLA-4criticalbindingsites.
