CD276(B7-H3)isamemberoftheB7/CD28superfamilyofcostimulatorymoleculesservingasanaccessorymodulatorofTcellresponse.B7familymolecules,whichareexpressedonantigen-presentingcellsanddisplayextracellularregionscontainingimmunoglobulin(Ig)variable(V)-andconstant(C)-likedomains,areknowntomodulateTcellreceptor(TCR)-mediatedTcellactivationbyprovidingco-signalsthatareeitherstimulatoryorinhibitory.B7-H3providesastimulatorysignaltoTcells.However,recentstudiessuggestanegativeregulatoryroleforB7-H3inTcellresponses.B7-H3inhibitedTcellproliferationmediatedbyantibodytoTcellreceptororallogeneicantigen-presentingcells.B7-H3isanegativeregulatorthatpreferentiallyaffectsT(H)1responses.B7-H3mayplayanimportantroleinmuscle-immuneinteractions,providingfurtherevidenceoftheactiveroleofmusclecellsinlocalimmunoregulatoryprocesses.Recently,B7-H3expressionhasalsobeenfoundinavarietyofdifferenthumancancers,includingprostatecancer,clearcellrenalcellcarcinoma(ccRCC),non-small-celllungcancer(NSCLC),pancreaticcancer,gastriccancer,ovariancancer,colorectalcancer(CRC)andurothelialcellcarcinoma.B7-H3wasexpressedinsomehumancancersandcorrelatedwithpooroutcomeofcancerpatients.
