CD27(TNFRSF7)isamemberoftheTNF-receptorsuperfamilylimitedtocellsofthelymphoidlineageandexistsasbothadimericglycoproteinonthecellsurfaceandasasolubleproteininserum.AsaTandBcellco-stimulatorymolecule,theactivityofCD27isgovernedbyitsTNF-likeligandCD70onlymphocytesanddendriticcells.TheCD27-CD70interactionisrequiredforTh1generationresponsestodifferentiationsignalsandlong-termmaintenanceofTcellimmunity,andmeanwhile,playsakeyroleinregulatingBcelldifferentiation,activationandimmunoglobulinsynthesis.TheCD27receptortransducessignalsandsubsequentlyleadstotheactivationofNF-κBandMAPK8/JNK,mediatedbytheadaptorproteinsTRAF2andTRAF5.Inaddition,theproapoptoticproteinSIVAiscapableofbindingthecytoplasmictailofCD27andexertsactionintheprocessofapoptosis.
