CD112(alsocalledNectin-2orpoliovirusreceptor-related2protein)isatypeItransmembraneglycoproteinandamemberoftheIggenesuperfamily(IGSF).CD112/Nectin-2functionsasanadhesionmoleculeinvolvedintheformationofcelljunctionsandinteractionswithotherNectin-familymolecules.Nectin-2isaalsoasurfaceMarkerofsomeendothelialcellsandregulatesmigration,proliferationandangiogenesis.CD112functionsasanimmunecheckpointregulatorbymodulatingTcellsignaling,eitherasacostimulatorofTcellfunction,orasacoinhibitorofTcellsproliferationbybindingtothereceptorsCD226orTIGIT(TcellIgandimmunoreceptortyrosine-basedinhibitorymotif(ITIM)domain),respectively.ThebindingofCD112ligandtothesereceptorsisweakcomparedtoCD155.Recently,anewreceptorofCD112,calledCD112RorPVRIG(Poliovirusreceptor-relatedimmunoglobulindomain-containingprotein)hasbeencharacterized,showinghighaffinitytoCD112.CD112RisanewcoinhibitoryreceptorofthePVRfamilyforhumanTcells,inhibitingTCR-mediatedsignalsuponbindingtoCD112/Nectin-2.CD112RispreferentiallyexpressedinTlymphocytesandNKcells.CancercellsevadeimmuneresponsesasaresultofanimmunosuppressivetumormicroenvironmentandCD112R,byplayingaroleintheregulationoftheimmunesystem,couldbeanewattractivecancerimmunotherapytarget.
