BreastCancer1(BRCA1)AssociatedProtein-1(BAP1)isadeubiquitinatingenzymethatwasidentifiedinascreenforproteinsthatinteractwithBRCA1.Thehumanproteinhasapredictedmolecularweightof81kDaandshares93%and92%aminoacid(aa)sequenceidentitywiththemouseandratorthologs,respectively.BAP1hastwofunctionaldomains,anN-terminalubiquitinC-terminalHydrolase(UCH)domainandaC-terminalBRCA1-interactingdomain.TheUCHdomaincontainsanactivesitecysteineresidue,Cys91inhumans,whichcatalyzesthehydrolysisofubiquitinisopeptidebonds.BAP1alsocontainstwoconsensusnuclearlocalizationsequences,NLS1,fromaa656-661,andNLS2,fromaa717-722.NLS2,butnotNLS1,isrequiredforthenuclearlocalizationofBAP1.BAP1formsaternarycomplexwithhostcellfactor1(HCF-1)andYY1andregulatestheexpressionofseveralgenes.DeubiquitinationofHCF-1byBAP1wasreportedtoberequiredforefficientG1/Sphasetransition,suggestingthatBAP1maybeinvolvedincellcycleregulation.AccumulatingevidenceindicatesthatBAP1alsoactsasatumorsuppressor.Forinstance,inactivationorlossofBAP1isassociatedwithseveraltypesofcancer,includingmyelodysplasticsyndrome,melanoma,renalcellcarcinomaandmalignantmesothelioma.
