BAFFismainlyproducedbyinnateimmunecellssuchasneutrophils,monocytes,macrophages,dendriticcells,folliculardendriticcells.Tcells,activatedBcells,somemalignantBcellsandalsonon-lymphoidcellslikeastrocytes,synoviocytesandepithelialcellscanalsoproduceBAFF.BAFFbindsthreedistinctreceptors(BAFF-R,TACIandBCMA)expressedpredominantlyonBcells,althoughactivatedTcellsalsoexpressBAFF-R.BAFFisamasterregulatorofperipheralBcellsurvival,andtogetherwithIL-6,promotesIgclass-switchingandplasmacelldifferentiation.BesidesitsmajorroleinBcellBIOLOGy,BAFFco-stimulatesactivatedTcells.DeregulatedexpressionofBAFFleadstoautoimmunedisordersinmice.Inhumans,elevatedlevelsofsolubleBAFFhavebeendetectedintheserumofpatientswithvariousautoimmunediseasessuchasSjoegrensyndrome,Rheumatoidarthritis(RA),Multiplesclerosis(MS)andSystemicLupusErythematosus(SLE).BAFFhasalsoincreasedlevelsinsomelymphoidcancers.
