The26SProteasomeisthemajornon-lysosomalproteaseineukaryoticcellsandisresponsIBLeforthedegradationofubiquitinatedsubstratesandmisfoldedproteins.Itiscomposedoftwosubcomplexes:the20SProteasomecoreparticleandthe19SProteasomeregulatoryparticle.The20SProteasomefacilitatesproteolyticcleavageofproteinsubstratesandiscomposedof28subunitsarrangedintofourstackedrings.Theouterringsofthe20SProteasomearecomposedofsevenrelatedbutnon-identical,non-catalyticsubunits,α1-7,thatformagateandrestrictsubstrateaccess.Theinnerringsofthe20SProteasomearecomposedofsevenrelatedbutnon-identicalsubunits,β1-7.β1,2,and5haveproteyolyticactivity.The19SProteasomecapsoneorbothendsofthecoreparticleandregulatessubstrateaccesstothecatalyticcoreinanATP-dependentmannerbymodulating20SProteasomeconformation.The19SProteasomeconsistsofabasesubcomplexandalidsubcomplex.ThebasesubcomplexiscomposedofsixAAA+familymembers,scaffoldingproteins,andregulatoryproteinsinvolvedinubiquitinrecognition.The19SProteasomelidsubcomplexcontainseightsubunitsplusonedeubiquitinatingenzyme,Rpn11.Smallmoleculesthatinhibittheactivityofthe26SProteasomeareusedtostudythefunctionofshort-livedintracellularproteinsandfortheclinicaltreatmentofcertainformsofcancer.
