Synucleinisasmall,solubleproteinprimarilyexpressedinneuraltissue.Thesynucleinfamilyincludesthreeknownproteins:α-synuclein,β-synuclein,andγ-synuclein.α-Synucleinisaveryconserved,smallacidicprotein(140residues)withamolecularweightofapproximately19kDathataccountsforabout1%oftotalproteininneurons.ItisconcentratedprimarilyinpresynapticaxonterminalsandplaysacentralroleininitiatingParkinson"sdisease.ItistheprincipalconstituentofLewybodieswhichareformedbyproteinaggregationandinclusionbodyformation.α-Synucleinlackssecondaryortertiarystructure,soitbelongstothefamilyofnativelyunfoldedproteins,manyofwhichactaschaperones.Muchofthisα-synucleinhasbeenpost-translationallymodifiedtoincreasetherateofoligomerization.Mutantformsofα-SynucleinareassociatedwithautosomaldominantParkinson"sdisease.Themutationsincreasetheaggregationrateoftheresultantα-synuclein.
