Zinc-α-2-glycoprotein(ZAG),firstidentifiedinthe1960s,derivesitsnamefromitsprecipitationfromhumanplasmaupontheadditionofzincsalts.ZAGhassincebeenfoundinsecretoryepithelialcellsandinarangeofbodyfluids.ZAGisidenticaltoalipidmobilizingfactorisolatedfromtheurineofpatientswithcancercachexiaandstimulateslipolysisininvitroandinvivoexperiments.Duetoitsexpressionin,andsecretionfromADIpocytes,ZAGisconsideredanadipokine.RecentlytheclinicalsignificanceofZAGhasbeenclarified.ZAGexpressioninadipocytesisinverselyrelatedtofatmass,thusitisintimatelyinvolvedinthemaintenanceofbodyweightinmiceandhumans.EpidemiologicalstudieshaveuncoveredanassociationbetweenZAGandplasmacholesterol.Thenon-synonymoussinglenucleotidepolymorphismrs4215inZAGisassociatedwithplasmacholesterolandobesity.StructurallyZAGpossessesaclassImajorhistocompatibilitycomplex(MHC)proteinfold.Itisdistinctfromothermembersofthisproteinfamilyinthatitissoluble,ratherthanbeinganchoredtoplasmamembranes,anditassociateswithprolactininducIBLeproteinratherthanβ2-microglobulin.Similartopeptideantigen-presentingclassIMHCmolecules,ZAGpossessesanopenapicalgroovebetweenitsα1andα2domainhelices.
