Interleukin-8(IL-8)wasoriginallydiscoveredasaneutrophilchemotacticandactivatingfactorandisamemberoftheα(CXC)subfamilyofchemokines(includingalsoplateletfactor4,GRO,IP-10,etc.).Manycelltypes,includingmonocyte/macrophages,Tcells,neutrophils,fibroblasts,endothelialcells,keratinocytes,hepatocytes,chondrocytesandvarioustumorcelllines,produceIL-8inresponsetoawidevarietyofproinflammatorystimulisuchasexposuretoIL-1,TNF,LPSandviruses.IL-8hasawiderangeofotherproinflammatoryeffects.Itisapotentchemoattractantforneutrophilsandcausesdegranulationofneutrophilspecificgranulesandazurophilicgranules.IL-8inducesexpressionofthecelladhesionmoleculesCD11/CD18andenhancestheadherenceofneutrophilstoendothelialcellsandsubendothelialmatrixproteins.Besidesneutrophils,IL-8isalsochemotacticforbasophils,Tcellsandeosinophils.IL-8hasbeenreportedtobeaco-mitogenforkeratinocytesandwasalsoshowntobeanautocrinegrowthfactorformelanomacells.IL-8wasalsoreportedtobeangiogenicbothinvivoandinvitro.
