Thekinasescalledp38MAPkinases(MAPKs)areintracellular,solubleserine-threoninekinaseswhichbelongtoalargefamilyofproteinsthatincludetheextracellularregulatedkinases(ERKs)andc-JunN-terminalkinases(JNKs).Sofarfourp38isoformshavebeenidentified,namelyp38α,p38β,p38γandp38δ,a.k.a.,p38-α(MAPK14),-β(MAPK11),-γ(MAPK12/ERK6),and-δ(MAPK13/SAPK4).Theubiquitouslyexpressedsp38αhasbeenthemostextensivelystudiedandisbelievedtobethemostphysiologicallyrelevantintheregulationoftheinflammatoryresponse.Theroleofthethreeotherisoformsisnotcurrentlywellunderstood,howevertheirprimarysitesofexpressionareknown.Similartop38α,p38βisalsoubiquitouslyexpressed,whilep38γisexpressedpredominatelyinskeletalmuscleandp38δisexpressedprimarilyinthelung,kidney,testis,smallintestineandpancreas.ThesmallmoleculeinhibitorVX-702isapotentp38kinasefamilyinhibitorthathasbeenstudiedforitseffectsoninflammationandtheinflammatoryresponse.VX-702dose-dependentlyinhibitedtheproductionoIL-6,IL-1βandTNFα(IC(50)=59,122and99ng/ml,respectively),andinanti-plateletaggregationassays,pre-incubationofplateletswithVX-702(1µM)completelyorpartiallyinhibitedplateletagoNISTinducedp38activation(IC(50)=4to20nM).MorerecentlyVX-702hasbeenstudiedasapotentialtreatmentforrheumatoidarthritis(RA).VX-702appearstobemosteffectiveagainstMAPK14,followedbyMAPK11andtheremainingmembersofthefamily,howeverexactinvitroIC(50)valueshavenotbeenpublished.HoweveralargeamountofdataisavailableforinvitrogrowthinhibitionassayswhereVX-702exhibitspoptentactivity,havinglowIC(50)valuesbeginningat~16nMandendingat<400µm=""for=""the=""numerous=""cell=""lines="">
