TMBZisachromogenicreagentutilizedforperoxidasedetection.Ithasbeendevelopedasanalternativetobenzidine.AlthoughtheTMBZsolutioniscolorless,itturnsbluish-green(λmax:655nm)inthepresenceofhydrogenperoxideandperoxidase.Thestructureofthisbluish-greencomplexisthoughttobearADIcalformoftwooxidizedTMBZmolecules.Thisperoxidasesubstrateisusedinstainingproceduresinimmunohistochemistryaswellasbeingavisualisingreagentusedinenzyme-linkedimmunosorbentassays(ELISA).Thesubstrateproducesasolubleendproductthatispaleblueincolorandcanbereadspectrophotometricallyat370or620-650nm.TheTMBreactionmaybestoppedwith2MH2SO4(resultinginayellowcolor),andreadat450nm.
