Long-wavelengthcell-permeantCa2+indicator.Rhod-2/AMisanacetoxymethylesterderivativeofRhod2thatcanbeeasilyloadedintocellsviaincubationandusedforquantifyingintracellularCa2+concentrationallowingnon-invasivemeasurementofcalciumionsinlivecells.ThisfluorogenicprobeforintracellularCa2+operatesinthevisIBLespectrum.Rhod-2/AMitselfdoesnotbindCa2+,butitisreADIlyhydrolyzedtoRhod-2byendogenousesterasesoncethedyeenterscells.Rhod-2isavaluablealternativetoFluo-3toinvestigatechangesinintracellularfreeCa2+concentrations,especiallyincellsandtissuesthathavehighlevelsofautofluorescence.Rhod-2hasthelongestfluorescentemissionsignalofthecommonlyusedcalciumindicators.Itcontainsarhodamine-likefluorophore,whoseexcitationandemissionmaximamakeitsuitableforusewithargonandkryptonlaser.Itssignalintensityisthestrongestofallthecalciumprobes.SpectralData:λex550nm,λem578nminmethanol;λex549nm,λem578nminDMSO;λex550nm,λem570nminChloroform:Methanol(90:10).
