Itisknownthatphenolandp-cresolarepromotersofskintumorsandalsourinaryexcretionofphenolandp-cresolisincreasedbyexposuretobenzene,whichhashemopoietictoxicity,andtoluene,respectively.Therefore,thedeterminationofphenolandp-cresolisimportantinclinicalchemistryandtoxicology.Becausephenolandp-cresolarepresentinurineassulfateorglucuronideconjugates,theyarehydrolyzedenzymaticallyorwithacid,followedbydistillationorextractionwithorganicsolventpriortomeasurementbyGCorHPLC.However,theseGCandHPLCmethodsrequirerelativelylargesamplevolumes(morethan1ml)becauseoftheirinsufficientsensitivitiesformeasuringsmallamountsofphenolandp-cresol.TheapplicationoffluorescentlabelingtechniquestoHPLCresultinasensitiveandselectivemethod.PhthalimidylbenzenesulfonylchlorideshabealsobeensuccessfullyusedforhighsensitivitylabellingofaminoacidspriortoHPLC.
