NADPH-diaphorasesubstratethatcompetitivelyinhibitsNOS(nitricoxidesynthase).Well-knownscavengerofsuperoxideanions.Dyethatisusedfordetectionofalkalinephosphataseincombinationwith5-bromo-4-chloro-3-indoxylphosphate(BCIP).ThissubstratesystemproducesaninsolubleNBTdiformazanendproductthatisblueincolorandcanbeobservedvisually.WhenusedwithBCIP,itissuitablefordetectionofalkalinephosphataseinwesternblots,forimmunohistologicalstainingproceduresandforcolorimetricindicationofbacterialinfectioninbloodsamples.Usedasaredoxindicatorforenzymaticreactionsincludingdehydrogenases,threoninedeaminase,glucose-6-phosphatedehydrogenase,phosphofructokinaseonpolyacrylamidegels,oxidasesonpolyacrylamidegelsandpentoseshuntdehydrogenses.TheNBT/BCIPreactionisalsousedforcolorimetric/spectrophotometricactivityassaysofoxidoreductases.Oneapplicationisinactivitystainsingelelectrophoresis,suchaswiththemitochondrialelectrontransportchaincomplexes.
