Uncharged,hydrophobic,photostablefluorescentprobethatstronglyfluorescesbrightredinhydrophobic(lipid-rich)environments,butisalmostnonfluorescentinwater.ThislipophilicstainiscommonlyusedforthedetectionofintracellularlipiddropletsincellssuchasADIpocytesbyfluorescencemicroscopyandflowcytometry.Ithasbeenshowntodetecttheexposureorformationofnewhydrophobicsurfacesinducedbyligandbindingtocalmodulin,oligomerizationofmelittin,orunfoldingofovalbuminduringearlythermaldenaturation.Thisproducthasbeenemployedtostaincellstructures,aswellastovisualizeandlocalizecolloidaldrugcarriers.HasbeenusedtostainlipophilicproteinsthathavebeenseparatedbySDS-PAGEelectrophoresisortodetectsphingolipidsonthin-layerchromatograms.ThefluorescenceisunaffectedbetweenpH4.5and8.5andmaybeusedindiluteconcentrationstoinvestigatehydrophobicproteinsites.Spectralproperties:Intracellularfatvacuoles,filledwithneutrallipidssuchascholesterol,lipoproteinsandtriglycerideswillfluorescegreen(Abs/Em=485/525)whilepolarlipids,suchasphospholipids,willfluorescered(Abs/Em=552/636nm).
