MAPTAMisacellpermeableintracellularCa2+chelatorthatcanbeloadednon-invasiveintocellsbyincubation.MAPTAMitselfdoesnotbindcalcium,butonceinsidethecellisconvertedintoit"sderivativeDimethyl-BAPTAbycytoplasmicesterases.Thistypeofcalciumchelatorsarecommonlyusedtoformcalciumbufferswithwell-definedcalciumconcentrations.ByinjectingthechelatorsintocellsorbyincubatingcellswiththeAMesterformofthechelators,onecancontrolthecytosoliccalciumconcentration,animportantmeanstostudytherolesofcalcium.KeyadvantagesofthesecalciumchelatorsincluderelativeinsensitivitytowardintracellularpHchangeandfastreleaseofcalcium.RecentstudieshaveshownthatBAPTAderivativemighthavemicrotubuledisruptiveactivity(unrelatedtoit"scalciumchelatingactivity)andshouldbeusedwithcautioninstudiesofCytoskeleton-relatedcellfunctions.
