Itisknownthatphenolandp-cresolarepromotersofskintumorsandalsourinaryexcretionofphenolandp-cresolisincreasedbyexposuretobenzene,whichhashemopoietictoxicity,andtoluene,respectively.Therefore,thedeterminationofphenolandp-cresolisimportantinclinicalchemistryandtoxicology.Becausephenolandp-cresolarepresentinurineassulfateorglucuronideconjugates,theyarehydrolyzedenzymaticallyorwithacid,followedbydistillationorextractionwithorganicsolventpriortomeasurementbyGCorHPLC.However,theseGCandHPLCmethodsrequirerelativelylargesamplevolumes(morethan1ml)becauseoftheirinsufficientsensitivitiesformeasuringsmallamountsofphenolandp-cresol.Inthegeneral,theapplicationoffluorescentlabelingtechniquestoHPLCresultinasensitiveandselectivemethod.Someacylchloridesareknowntoreactwithphenolichydroxygrouptoformesters.Itisknownthat4-(N-phthalimidinyl)benzensulfonylchloridePhisyl-Cl7,havingsulfonylchloride,reactedwithphenoltogivethecorrespondinghighlyfluorescentsulfonylester.ThesensitivityofPhysil-ClwascomparedwithDansyl-ClandDPS-Cl,whichareanalogoustoPhysil-Clasfluorescentlabelingreagentshavingsulfonylchloridegroup.ThelabelingreactionofphenolwithDansyl-ClandDPS-ClwerecarriedoutaccordingtothemethodofRuiteretal.5andthepeaksweredetectedattheeachfluorescencemaximawavelength(Ex350nmaEm550nmforDansyl-Cl,andEx320nmaEm386nmforDPS-Cl).PeakheightofPhisylderivativewashigherabout85-and7,3-foldthanthosseofDansylderivativeandDPSderivative,respectively.Amongthesethreereagents,Phisyl-Clwasthemostsensitive.
