JC-10issuperioranalogofJC-1,usefulfordeterminingmitochondrialmembranepotential(MMP)incellsbyflowcytometry,fluorescencemicroscopyandinmicroplate-basedfluorescentassays.JC-10isacationic,lipophilicdyethataccumulatesinmitochondriaofcellswithapolarizedmitochondrialmembrane,selectivelygeneratinganorangeJ-aggregateemissionprofile(λex=540nm/λem=590nm)inhealthycells.Uponcellinjuryorcelldeath,asmembranepotentialdecreases,whichresultsinthefailuretoretainJC-10aggregatesinthemitochondriaandJC-10monomersaregeneratedresultinginashifttogreenemission(λex=490nm/λem=525nm).JC-10showsimprovedsolubilitycomparedtoJC-1inaqueousmediaandanabilitytodetectsubtlerchangesinmitochondrialmembranepotentialloss.JC-10allowsforqualitativevisualization(shiftfromorangetogreenfluorescence)andquantitativedetection(fluorescenceintensityratio)ofmitochondrialmembranepotentialchanges.WavelengthMaxima:λex=510nm|λem=525nm.
