Inrecentyearsseveralgroupshavestudiedthesequencerequirements,specificity,signallingpathwaysandkineticsoftheTLR(Toll-likereceptor)9suppressionbyinhibitoryoligonucleotidemotifs,whichledtoaclassofnovelinhibitoryoligonucleotide(iODNs),thatisindependentofthepreviouslythoughtspeciespreference.SubsequentlyithasbeendiscoveredthattelomericDNArepeats(TTAGGG)ncanblockimmuneactivationbyCpG-ODNs.Short,11-15baselongoligonucleotidesweresynthesizedthatwerecapableofpotentlyinhibitingCpG-stimulation.TheoptimalinhibitoryDNAmotifconsistsofapyrimidine-richtriplet,preferablyCCT,whichispositioned5-totheGGGsequenceinasinglestrandedDNAmolecule.Additionally,boththeoptimalspacingbetweentheCCTandGGGmotifs,aswellastheirrelativeordertoeachother,isofcrucialimportancefortheinhibitoryDNAaction.Interestingly,althoughbothTLR7/TLR8ligandsandbacterialDNAsharetheendosomalcompartmentforreceptorbindingandsignaltransduction,certainiODNs(G-type)suppressonlyTLR9-mediatedactivation,whereasprototypeclassIiODNmayalsointerferewiththeactivationviatheTLR7/TLR8pathway.Recently,intriguingevidencehasbeenpresentedthatforsomeiODNclassestheimmuno-modulatoryBIOLOGicalactivityshowsonlylimitedsequencedependencyormaynoteveninvolveTLR-mediateduptakeandsignalingpathways.ForexampleiODNsoftheclassIIarethoughttoactonimmuneactivationthroughinhibitionofSTATsignallingandindependentofTLRsignallingviabindingtoayettobeidentified"ODN-receptor".SlightlymodifiedphosphodiesterversionsofthemostpotentinhibitoryODNswerealsoabletoprofoundlyblocktheimmuneactivationofmacrophagesandjustrecentlyprovetobevaluabletoolsforinvivouseinexperimentalanimalmodelsofinflammatoryandauto-immunediseases.BasedupontheserecentinsightsthefollowingclassificationforiODNshasbeensuggested:
